In female mammals, dosage compensation for X-linked genes is ensured through random X-chromosome inactivation mediated by the long non-coding RNA Xist. During this process most genes are silenced on one randomly chosen X-chromosome. In embryonic stem cells however, where both X-chromosomes are active, double X-dosage affects pluripotency and differentiation through inhibiting the MAPK signaling pathway. We combine theoretical and experimental approaches to understand how X-dosage information will drive cellular decision making processes.
